Bacillus subtilis is a species of Bacillus, CAS number 68038-70-0. A Single cell is 0.7-0.8×2-3 microns, uniformly colored. No capsule, flagella around, can move. Gram-positive bacteria can form endogenous stress-resistant spores, spores are 0.6-0.9×1.0-1.5 microns, elliptical to columnar, located in the center of the bacteria or slightly off, and the bacteria do not swell after spore formation. Growth and reproduction speed is fast, the colony surface is rough and opaque, dirty white or slightly yellow, and wrinkles are often formed when growing in a liquid culture medium. It is an aerobic bacterium.
It can use protein, various sugars, and starch to decompose tryptophan to form indole. It is widely used in genetic research, and the synthesis pathway of purine nucleotides and its regulatory mechanism of this bacterium is well studied. It is widely distributed in soil and corrupted organic matter and is easy to reproduce in the juice of dead grass. Some strains are important producers of α-amylase and neutral protease; some strains have enzyme systems that strongly degrade nucleotides, so they are often used as parent strains for breeding nucleoside-producing bacteria or for producing 5'-nucleotidase.
Main characteristics:
Bacillus subtilis is a species of Bacillus, widely distributed in soil and decayed organic matter, and is named for its easy reproduction in the juice of dead grass. The single cell of this bacterium is (0.7~0.8)μm×(2~3)μm, and is evenly colored. It has no capsule, or flagella, and is active. Gram staining is positive. The spores are (0.6~0.9)μm×(1.0~1.5)μm, located in the center of the bacterial body or slightly off, and the bacterial body does not swell after spore formation. The colony's surface is rough and opaque, dirty white or slightly yellow, and often forms wrinkled walls when growing in a liquid culture medium. It is an aerobic bacterium. It can use protein and various sugars to decompose tryptophan. Among them, Bacillus subtilis has great potential in biological control. Under suitable conditions such as temperature and ventilation, a large number of spores are formed in young cells. In addition, B. subtilis grows fast, has low nutritional requirements, can efficiently secrete many proteins and metabolites, and does not produce toxins. It is a non-pathogenic and safe microorganism. It has a wide range of uses in medicine, health, food, etc. In microbial experiments, the culture medium formula commonly used for detecting and observing Bacillus subtilis is 1 L distilled water + 20 g glucose + 15 g peptone + 5 g sodium chloride + 0.5 g beef extract + 20 g agar.
Bacillus subtilis has two growth periods: spore dormancy period and reproductive growth period. B. subtilis will enter the spore dormancy period in unsuitable environments such as harsh growth environments and lack of nutrients, and form spores with extremely strong stress resistance and can survive in polar environments such as high temperature, acid, and alkali, to adapt to the environment and survive. Once the environment becomes suitable for growth and nutrition is sufficient, the spores will automatically enter the reproductive growth period and grow back into Bacillus subtilis.
